Ciliate protozoa living in the intestine of the great apes

Ushida Kazunari

13th November,2006 - 1st December,2006

Fresh feces were collected from several chimpanzees in Tacugama Chimpanzee Sanctuary. A portion of feces was diluted with 4 weights of Phosphate buffered saline (PBS) previously warmed to ca. 37oC..Mixture was squeezed through two layers of surgical gauze and added 1g of sucorose or glucose. Transfer filtrate to a sepqration funnel of 100-ml and placed in a tap water of ca. 37oC. Separation funnel stand for 1 hr to sediment the ciliate protozoa. The lowest layer was recovered and quickly fixed in MSF solution or EtOH. Before fixation, Trogrodytella spp. were microscopically checked. This smaple collection was repeated for three times during the stay.Fresh feces were individually collected for the detection and identification of Bifidobacteria. Feces were fixed in EtOH for preservation of bacterial DNA. After the sample collection in Tacugama chimpanzee Sanctuary, I moved to Conakry to attend the Bossou 30ans symposium co-organized by KUPRI, DNRST, and IREB
